Discuss the basic requirements and working process of setting up microbial sterility room
Asepsis room, also known as vaccination room, is a special laboratory for systematic sterilization, sterilization and other aseptic operations. In microbial work, inoculation and transplantation of bacteria is a major operation. The characteristics of this operation are to ensure the pure strain of bacteria and prevent the contamination of miscellaneous bacteria. In the air of general environment, because there are many dust and bacteria, it is easy to cause pollution, which greatly interferes with the vaccination work.
The establishment of the system should be based on the principles of economy and science. Its basic requirements are as follows:
(1) there should be two inside and outside, and the other is a buffer room. Room volume should not be too large to facilitate air sterilization. The interior area is 2 *2.5=5m2, the exterior area is 1 *2=2m2, and the height is below 2.5m. There should be a ceiling.
(2) Sliding doors should be installed in the interior to reduce the fluctuation of air. The doors should be located far from the workbench zui. The exterior doors should also be located far from the interior zui.
(3) A small window should be opened on the walls or "partitions" between the interior and the exterior so as to provide the necessary passageway for the delivery of goods inside and outside during the inoculation process, so as to reduce the number of people entering and leaving the interior and reduce the pollution level. The small window is 60 cm wide, 40 cm high and 30 cm thick, and the inside and outside windows are hung with opposite pull-out fans.
(4) the volume is small and tight. After a period of time, the indoor temperature is very high. Therefore, ventilation windows should be set up. The ventilation window should be set on the ceiling at the entrance door of the inner chamber (i.e. far from the worktable zui). The Zui is a double-layer structure, with the outer shutter and the inner shutter. The ventilation window can be opened after use in the inner room and before sterilization to circulate air. Sterile indoor equipment and appliances:
(1) the worktable in sterile room requires smooth surface and table level regardless of material and purpose.
(2) Install an ultraviolet lamp (mostly 30W) inside and outside. The indoor ultraviolet lamp should be installed directly above the regular working seat, 2 m above the ground. The outdoor ultraviolet lamp can be installed in the center of the outdoor.
(3) Outdoor work clothes, shoes, hats, masks, ceramic basins and towels containing Lazur water, hand-held sprayers and 5% carbolic acid solution should be specially designed.
(4) There should be alcohol lamps, commonly used inoculation tools, stainless steel knives, scissors, tweezers, 70% alcohol cotton balls, industrial alcohol, glass sheets, special crayons, notebooks, pencils, label paper, glue, waste baskets and so on.
Sterilization:
(1) fumigation: This is a thorough sterilization. It should be fumigated and sterilized for a long time when the pollution is serious. It can be fumigated with formaldehyde, lactic acid or sulphur.
(2) spray: before each use. Spraying can promote the deposition of particulates and microorganisms in the air, prevent dust flying field on the desktop and ground, and have bactericidal effect. It can be sprayed with 5% carbolic acid.
(3) ultraviolet radiation: before each use. Ultraviolet radiation has better bactericidal effect. Usually the ultraviolet lamp should be turned on for 30 to 60 minutes.
Working procedure:
(1) asepsis room sterilization. Ultraviolet light is turned on for more than 30 minutes before each use, or 5% carbolic acid spray is applied to the inner and outer rooms 30 minutes before use.
(2) After washing hands with soap, move the necessary equipment into the outdoor; change the sterilized overalls, hats and shoes into the outdoor, wear a mask, and then soak hands with 2% coal phenol soap for 2 minutes.
(3) Take all kinds of necessary articles into the inner room to count and put them in place. Spray 5% carbolic acid on the working table and the space of the operator's station. Return to the outer room and work in the inner room after 5-10 minutes.
(4) Before inoculation operation, wipe hands with 70% alcohol cotton ball; during aseptic operation, the movement should be gentle to minimize air fluctuation and dust on the ground.
(5) safety should be paid attention to in work. If the cotton plug is on fire, hold it tightly by hand or wrap it with wet cloth to extinguish it, do not blow it with your mouth to avoid enlarging the combustion; if the bacterial culture is sprinkled or broken, wrap it with a cloth soaked with 5% carbolic acid, wipe the table or floor with a cloth soaked with 5% carbolic acid, wipe hands with an alcohol asbestos ball and then continue to operate.
(6) at the end of the work, the table will be cleaned up immediately, and all the articles and wastes stored in the store should not be taken out. 30min will be irradiated with 5% carbon dioxide spray or ultraviolet lamp.
