Laminar flow static (empty state) air sampling method:
(1) When the air supply outlet is centralized, the operation area and the surrounding area should be detected separately; when the air supply outlet is distributed, the detection should be carried out according to the method of uniform distribution of the whole room.
(2) When sampling by sedimentation method, the culture dish with a diameter of 9 cm was exposed to air for 30 minutes and incubated at 37 (?) for 24 hours. The number of colonies in the culture dish (cfu) represented the number of bacteria that settled in the air, expressed as a number of dishes.
(3) When sampling by plankton method, the fissure sampler is used to sample in air for 30 minutes at random. The number of colonies (cfu) cultured from sampling dishes or strips represents the number of bacteria floating in the air, expressed as one/m3.
Laminar flow static (empty state) sampling height:
(1) the sampling points may be placed on the ground or not at any height above the ground 0.8m.
(2) there should be no operating table in the operation area; when the operating table is fixed, the detection height should be 0.25cm above the table.
Notes on laminar flow static (empty state) sampling:
(1) In the 100-level area detection, the sampling port should be in the direction of the airflow; in other areas, the sampling port should be upward;
(2) Sampled medium strips or dishes should be cultured for 24 hours at 37 C, and the number of colonies should be calculated. The average number of colonies should be rounded to one decimal place.
(3) When the concentration of planktonic bacteria is measured by plankton method, the number of bacterial concentration measurement points should be the same as that of dust concentration measurement points in the measured area, and should be in the same position. Each sampling should meet the minimum sampling requirement of Table 8, and each sampling time should not be less than 30 minutes.
(4) When the sedimentation method is used to determine the concentration of sedimentation bacteria, the number of bacterial concentration measurement points is no less than the number of dust concentration measurement points, which should meet the minimum number of Petri dishes specified in Table 9 (without reference dishes). If the settling time is prolonged properly, the minimum number of Petri dishes can be reduced proportionally, but not less than the minimum number of measuring points of dust concentration.
(5) No matter what method is used to detect bacterial concentration, there must be two blank controls. For the first time, it was used to detect Petri dishes or culture medium strips (one control dish per batch); for the second time, it was used to control the operation process (one control dish per room or area was opened and immediately sealed for simulated operation); for the second time, the control results must be negative, and the whole operation process should meet the requirements of aseptic operation.
(6) When accepting a new house, the bacteria on the air and surface should be tested at last.
